The Health Risks Of Phenethylamines

Nevertheless, the three-lever procedure provided “proof of concept” and was the first to show that animals could be trained to discriminate the stimulus effects of optical isomers of a given agent. There is only one phenethylamine that is a classic psychedelic, which is mescaline. MDMA and other novel designer psychostimulants often include serotonergic pharmacology and many act to increase serotonin or bind ‘psychedelic’ 5HT2A receptors. These substances often have considerable pharmacology at other neurotransmitter systems such as dopamine and norepinephrine. These drugs typically release serotonin more readily than norepinephrine or dopamine and are thought to provide a hybrid of psychostimulant and psychedelic properties. MDMA and many others may be arguably better classified as enactogens ‘to touch within’ or empathogens ‘generating a state of empathy’ than psychedelics ‘mind-manifesting’.
A substituted phenyl group at the Ar position greatly reduced the DA reuptake inhibitory activities of the compounds compared to those of the others with identical substitutions at R1 and R2 (comparing compound 2vs.3 and 4; compound 6vs.7 and 8). Compounds with a methoxy group at the Ar position showed very weak or no DA reuptake inhibitory activities (compounds 7, 8, and 12-18). Changes in the substituents at R2 from amino to aminomethyl groups produced the opposite effects, depending on the nature of the substituent at the Ar position (compound 1vs.5; compound 2vs.6). Even the well-known research work of the Shulgins42, which describes in detail phenethylamines and their physical properties, personal dosages used, duration of effects observed, and commentary on effects, does not contain information on these compounds. However, all three of the compounds studied here experimentally, 25B-NBF, 25C-NBF, and DMBMPP, are derivatives of the phenethylamine psychedelic 2C-B, which is indicated to have psychedelic effect at dosages between 12 and 24 mg42. Even so, this is not easily translatable to the concentrations used in our experiments due to the need to account for the mode of administration on distribution throughout the body. Food training and catheter implantation surgery were performed as described previously 48. The timeline for self-administration procedures is illustrated in Figure 4A. In brief, rats were trained to lever press for 45 mg food pellets (BioServ, Frenchtown, NJ, USA) under a continuous FR1 schedule of reinforcement during 1 h sessions. The distal end of the catheter was threaded subcutaneously and connected to a 22-gauge stainless steel cannula (P1 Technologies, Roanoke, VA, USA), fixed to the head assembly with dental cement and secured with Prolene surgical mesh (Ethicon Inc. Somerville, NJ, USA). After catheter implantation surgery, rats were allowed to recover from surgery for 7 days prior to the self-administration test.

To determine whether the β-PEAs that inhibited the DAT activity altered the functional activity of D2R, we selected two β-PEAs and determined their effects on the endocytosis of D2R. Dopamine transporter (DAT) plays a key role in terminating DA neurotransmission by reuptaking DA released into synapses (Carroll et al., 1999; Enyedy et al., 2003). DAT belongs to the solute carrier superfamily and is characterized by twelve transmembrane domains. It reuptakes synaptic DA released from presynaptic neurons by utilizing the energy of the sodium gradient generated by Na+/K+-ATPase (Kilty et al., 1991; Shimada et al., 1991).

For example, amphetamine increased 50-kHz USVs, which were inhibited by the blockade of the DAD1R antagonist SCH23390 in rats 52. Consistently, our results demonstrated that acute β-PEA administration at a dose of 50 mg/kg significantly increased the frequency of 50-kHz USV calls.
